LNP-mRNA Ultrafiltration Technology Showdown: TFF Cassette vs. Hollow Fiber Filter - A Case Study Efficient purification of lipid nanoparticles (LNPs) is critical for the development and manufacturing of mRNA-based therapeutics. As the "golden carrier" for nucleic acid delivery, LNPs require a robust downstream ultrafiltration process that preserves particle integrity while enabling high-purity concentration and buffer exchange. In this study, Cobetter evaluated two mainstream tangential flow filtration (TFF) formats—RC (regenerated cellulose) TFF cassettes and mPES (modified polyethersulfone) hollow fiber filters—for mRNA-LNP purification. Results showed that RC TFF cassettes outperformed in flux rate and overall processing efficiency, reducing membrane area and filtration time without compromising LNP attributes such as size, polydispersity (PDI), and encapsulation efficiency. In contrast, hollow fiber filters offered lower shear environments, making them suitable for shear-sensitive materials but exhibited relatively lower flux. Ultimately, both formats remain viable depending on specific material characteristics and process goals.
