Protein stability – all you need to know

Protein stability refers to the ability of a protein molecule to maintain its native conformation and functional activity under various environmental conditions, such as temperature, pH, and the presence of denaturing agents. It is an important aspect in pharmaceutical manufacturing, as the proteins (and resulting drug substances) derived from elaborate production processes must preserve their structural integrity until they have accomplished the task they were designed for.

Ensuring protein stability is essential not only for basic research in biochemistry but also for various applications in biotechnology, pharmaceuticals, and medicine. By elucidating the factors that influence protein stability and developing robust assays for its assessment, researchers can better design and optimize therapeutic proteins, improve protein engineering strategies, and enhance the storage and handling of protein samples.

In the course of this article, we will not only provide a brief overview of what protein stability is, but also discuss factors that have an impact on protein structure. Furthermore, we will highlight methods to assess protein stability, along with optimization opportunities for overall stability throughout protein manufacturing.
