This document presents a structural bioinformatics analysis of the insulin receptor family ectodomain based on phylogenetic information. The analysis confirms the location of ligand binding site 1 for the IGF1R and IR. It also predicts the potential location of insulin binding site 2 on the surface of the fibronectin type III domains of the IR. The analysis could not identify a conserved surface on the second leucine-rich domain that may interact with the ligand. The analysis suggests a possible mechanical trigger of IR activation involving a slight rotation of the last two fibronectin type III domains. It also finds strong selective pressure amongst IRR orthologous sequences, suggesting this orphan receptor has an unknown conserved physiological role.
