HISTROY
In year between 1876 AND 1884 ROBERT KOCH was the
first scientist to focus on isolation of bacteria in pure
culture
 This will give rise to ‘Germ theory’.
 According to this a certain micro organisms is
responsible for certain diseases .
ISOLATION :- IN MICROBIOLOGY ISOLATION REFERS TO
GETTING A PURE CULTURE.
 There are three basically methods of Isolation :-
 (i)streak plate
 (ii)spread plate
 (iii)pour plate
 STREAK PLATE
THEORY:- This is essentially a method to dilute
the number of organisms, decreasing the density -
individual colonies to be isolated from other
colonies. Each colony is "pure," since theoretically,
the colony began with an individual cell
 1.) Flame the loop, cool by touching an
uninoculated portion of the surFace.
2.) Now rotate the plate. Open lid and streak
again, remember: you are picking up growth
from quadrant one, and using this as your
inoculum for quadrant two.
3.) Flame loop; rotate plate, and repeat
procedure for quadrants three and four.
Methods of microbiolgy
This is an example of a
good streak for isolation
using the "four corners"
method.
(II) SPREAD PLATE
 THEORY :- A serailly diluted inoculum is
transferred over the surface of the solid medium
and then spreaded uniformly with a sterile bent
glass spreader.
 Colonies can be easily isolated from this method.

POUR PLATE
THEORY:- In pour plate , a small amount of culture is added by
pipette to the cenre of petri dish .cooled ,but molten agar medium in a
test tube or bottle is then poured in petridish . The dish is then
rotated gently ,to ensure the culture is mixed properly
