Non-compartment model or analysis, also called as
the model independent method.
This approach based on the Statistical moments
theory.

What is Non-compartment model?
• Non-compartmental method used to calculate
pharmacokinetic parameters. It does not assume
the number of compartments. It assumes that
elimination follows first order. This method is
simple and used commonly to calculate
pharmacokinetic parameters such as half-life,
clearance, and volume of distribution.

Why do we need Non-
compartment model?
• If the primary requirement is to determine the
degree of exposure following administration of a
drug (such as AUC), and the drug's associated
pharmacokinetic parameters, such as clearance,
elimination half-life, T (max), C (max), etc., then
Non-compartmental analysis is generally the
preferred methodology to use in that it requires
fewer assumptions than model-based
approaches.

As per Statistical moments theory or Non-
compartmental analysis:

Where MRT = mean residence time
AUMC = area under the first moment curve
AUC = area under the zero moment curve
Mean residence time (MRT) is the average time that the drug stays
in the body (or plasma as measured).

-AUMC is obtained from a plot of product of plasma drug
concentration (C) and time (t), i.e. C.t vs time (t) from zero to infinity.
Mathematically it is expressed as:

The AUMC & AUC can be calculated from the given
respective graphs or plots by the “trapzoidal rule.”
(Trapezoidal method involves dividing curve by a series
of vertical lines into a number of trapezoids {flat shape
with 4 sides} calculating separately the area of each
trapezoid and adding them together).
