This paper presents a novel optical flow-based framework for analyzing leukocyte dynamics in microcirculation via intravital microscopy. It employs a duality-based total variation L1 optical flow approach that effectively tracks leukocyte behavior while addressing illumination changes and outliers, and aims to provide precise hydrodynamic parameters. The proposed method enhances the automation of leukocyte detection, potentially advancing our understanding of inflammatory processes and aiding in drug development.
