Editor's Notes

  • #2: Does your next experiment involve CSF3? This video is for scientists who want to run Western blot, IHC, flow or other immunoassays against CSF3.
  • #3: We will cover some basic facts, protein expression, and CSF3 expected behavior in Western blot. All information in this video is based on public information and no proprietary experimental evidence was used.
  • #4: Topic 1: Quick facts about CSF3. Granulocyte colony-stimulating factor, or CSF3, is a cytokine that acts in hematopoiesis. It controls the production, differentiation, and function of two types of white blood cells, granulocytes and monocytes or macrophages. The CSF3 protein is 207 amino acids long and runs at about 22 kilodaltons on Western blot.
  • #5: Topic 2: Where is CSF3 expressed? High levels of CSF3 RNA have been found in the respiratory system and the bladder, with the highest levels detected in the lungs. It is also expressed at lower levels in the male and female tissues, muscles, connective tissues, and bone marrow. The protein is secreted and released outside the cell membrane. You can find more information on protein atlas dot org to check out which cell lines express CSF3 and which ones don’t. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
  • #6: Topic 3: CSF3 in Western blotting. Antibody specificity can make or break an experiment. After looking through Western blot images from publications and antibody companies, we can see a single band in a CSF3 Western blot at around 23 kilodaltons. Check out the PMC link for more images, which is also provided in the description box.
  • #7: Topic 4: Some interesting facts about CSF3. G-CSF or CSF3 is a member of the IL6 superfamily of cytokines that regulates production, differentiation, and function of granulocytes and monocytes. It was first recognized and purified in 1983 and was cloned in 1986. Scientists have explored the potential of CSF3 in treating neutropenia, a disorder characterized by an extremely low number of neutrophils in blood.
