  • #4: Topic 1: Quick facts about Periostin. Periostin is an extracellular matrix protein that is involved in wound healing. The protein is 836 amino acids long and runs about 93 kilodaltons on a western blot.
  • #5: Topic 2: Where is Periostin? Periostin is expressed in the skin, gastrointestinal tract, liver and gallbladder, kidney, urinary tracts and other tissues. It is also expressed on the periosteum, which is a type of membrane on bones. You can find more information on protein atlas dot org to check out which cell lines express Periostin and which ones don’t. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
  • #6: Topic 3: Periostin in Western blotting. Antibody specificity can make or break an experiment. After looking through western blot images from publications and antibody companies, we can see that typically one band can be expected in a Periostin western blot at about 93 kilodaltons. Since then, it has been shown to overexpress in certain cancers, such as ovarian, breast, colorectal cancer, and other types of cancer.
