Micropropagation and its stages:
Stage 0 selection of mother plant and its maintenance
stage I initiation and establishment of culture
Stage II multiplication of shoot or rapid somatic embryo formation
Stage III invitro rooting of shoot or germination of somatic embryo
Stage IV transfer of platelets to sterilized soil for hardening
• Micropropagation is the artificial process of
producing plants vegetatively through tissue
culture or cell culture techniques.
• Transfer and establishment of whole plants in
soil (HARDENING) is one the stages of
micropropagation.
Stages involved in hardening
HARDENING
• Hardening is gradual exposure of plantlets to an environmental conditions.
• Challenges faced by micropropagation:
a) Unable to compete with soil microbes and to cope with the environmental conditions
b) Altered morphology, anatomy and physiology.
• Agenda behind hardening in micro-propagation:
a) increase growth and reduce mortality in plantlets
b) efforts are focused on the control of both physical and chemical environment
c) bio-hardening of micro-propagated plantlets.
ACCLIMATIZATION
• Acclimatization: process of gradual hardening of the cultured plants from laboratory to field so that the regenerated
plants should adjust to field conditions.
• In this stage, the plantlets are established in the soil.
• The shoots from the laboratory are transferred to a greenhouse under controlled conditions of temperature, humidity and
light.
• The ultimate success of micropropagation on a commercial scale depends on the ability to transfer plants out of culture
on:
a) a large scale
b) at low cost
c) with high survival rates.
