  • #4: Topic 1: Quick facts about IL6. Interleukin-6 is a cytokine with a wide variety of biological functions in immunity, hematopoiesis, tissue regeneration, and metabolism. IL6 is primarily produced at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through IL6 receptor alpha. Rapid production of interleukin 6 contributes to host defense during infection and tissue injury, but excessive IL6 synthesis is involved in disease pathology. Interleukin 6 is 212 amino acids long and runs about 24 kilodaltons on Western blot.
  • #5: Topic 2: Where is interleukin 6 expressed? High RNA expression levels of IL6 is found in the brain, lungs, adipose tissue, bone marrow, and skeletal muscle cells, with the highest levels detected in the urinary bladder. They are also expressed at lower levels in the kidney, liver, thyroid gland, pancreas, skin, prostate and retina. Elevated levels of the encoded protein have been found in virus infections, including COVID-19. You can find more information on protein atlas dot org to check out which cell lines express interleukin 6 and which ones don’t. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
  • #6: Topic 3: Interleukin 6 in Western blotting. Antibody specificity can make or break an experiment. After looking through Western blot images from publications and antibody companies, we can see that typically one band at around 24 kilodaltons can be expected in an IL6 Western blot. Check out the PMC link for more images, which is also provided in the description box.
  • #7: Topic 4: Some interesting facts about IL6. Interleukin 6 was first discovered in 1986 as a T cell-derived lymphokine that induces the final maturation of B cells into antibody-producing cells. IL6 can simultaneously elicit distinct or even contradictory physiopathological processes. When taking into account all the accumulated basic research on the various aspects of this molecule, therapeutic strategies to inhibit IL6 signaling have proven effective in treating experimental models of autoimmune and chronic inflammatory diseases.
